Reliable detection of rare antigen-specific T and/or B cells has been a major challenge for immune monitoring. For over two decades, CTL has been leading this field with its ImmunoSpot® platform.

ImmunoSpot® test kits have not only been optimized for maximal sensitivity to detect and analyze the secretory footprint of individual T and B cells, but also for the unambiguous study of co-expression patterns.

CD8+ T cell subpopulations can be identified using a 4 color ImmunoSpot® kit that measures IFN-γ, TNF-α, GzB and IL-2 simultaneously (Learn more). See kit.

CD4+ T cell subpopulations Th1, Th2, Th17 and Th21 can be detected via their production of IFN-γ, IL-4, IL-17 and IL-21, respectively. Multiplex analysis of these subpopulations needs to account for their differential activation kinetics, however (learn more). As positive controls and reference samples, CTL offers PBMC/antigen pairs that display Th1, Th2, Th17 and Th21 reactivity (please inquire).

Up to 4 color kits are available for multiplexed detection of B cells secreting various immunoglobulin classes and/or IgG subclasses. Additionally, we will gladly assist you in developing antigen-specific B cell assays of your choice by formulating suitable coating conditions (please inquire).

All of our kits have been developed and standardized to eliminate the variability and subjectivity in performance results. Serum-free assay medium has been added within T-cell kits, eliminating the need for blocking and eliminating batch to batch variability seen in FBS-containing medium when used in functional assays.

Our ImmunoSpot® ELISPOT Kits include all buffers and washing solutions, and are available with our serum-free CTL-Test™ Medium that has been formulated for low background and- high antigen-specific signal performance in T cell ImmunoSpot® assays. CTL Serum-free Media products are also available for freezing and thawing of cryopreserved PBMC.

To complement our ImmunoSpot® kits, CTL also offers peripheral blood mononuclear cells (PBMC) with pre-defined properties for assay development. Such samples serve as invaluable positive controls and reference samples for assay validation and include PBMC that exhibit Th1/Th2/Th17 cytokine signatures or antibody-secreting cell (ASC) reactivity against defined antigen.
